Molecular Oncology Almanac (MOAlmanac) is an open-source precision oncology knowledge base jointly developed and maintained by the Broad Institute and Dana-Farber Cancer Institute in the United States. Its core goal is to organize the correlations between tumor molecular alterations and treatment sensitivity, drug resistance, and prognosis, providing structured high-quality reference data for oncology research. The current website mainly offers browsing and access to its underlying knowledge base, and all code and data of the entire project are openly hosted on GitHub.
To date, MOAlmanac has collected 1015 clinically relevant assertions corresponding to 153 molecular features. It supports multi-dimensional filtering by 73 cancer types, 6 evidence levels, and 179 therapeutic drugs, and allows Boolean logic combination queries. Search results are directly linked to corresponding literature, FDA approval information, or clinical guidelines. Data can be browsed online, downloaded as a complete TSV file, accessed programmatically via an API. It also comes with a Chrome extension and an analysis entry on the Terra cloud platform. The entire service is completely free, licensed under the Open Database License (ODbL), and only prohibits commercial use and clinical diagnostic use.
On the plus side, all content in this knowledge base is manually curated by professionals, with clearly classified evidence levels and traceable sources; it is freely open with multiple access methods to adapt to different research needs; the open-source license allows secondary development, making it highly flexible. The drawbacks are equally clear: it only supports research use and prohibits clinical use; it only has an English interface with no Chinese localization; its interaction design is academia-focused, unfriendly to non-professional users, and there is no commercial technical support.
